How It Works

Principle: Rich peptones and a phosphate buffer promote coliform growth, while lauryl sulfate suppresses Gram-positive bacteria. The chromogenic substrate X-GAL turns broth blue-green for coliforms, and the fluorogenic MUG glows blue under UV light for E. coli.
Medium: Cream to light yellow powder; 1.7% solution, soluble in water; pH 6.8 ± 0.2 at 25°C.

Procedure

Add the contents of one vial to 100 mL of water sample in a sterile bottle.
Incubate at 35-37°C for 18-24 hours (or 48 hours at 20-25°C).

Check results:

Negative: No color change (remains Yellowish).
Total Coliforms: Blue-Green color (X-GAL reaction).
E. coli: Blue-Green with UV fluorescence (MUG reaction).
