Principle of Use
Agarose consists of repeating galactose-based polysaccharide units that form a thermoreversible gel when heated in aqueous buffer and cooled. The resulting porous gel matrix allows nucleic acids to migrate under an electric field according to fragment size.
Pore size can be adjusted by concentration:

Higher concentrations (e.g. 2.0%) → smaller pores for small DNA fragments

Lower concentrations (e.g. 0.8%) → larger pores for high-molecular-weight DNA
Instructions for Use

Weigh the required amount of agarose powder

Suspend in appropriate electrophoresis buffer (TAE or TBE)

Heat until fully dissolved

Cool slightly and pour into a gel casting tray

Allow to solidify before loading samples
