Overview

BPLS Agar (Brilliant Green Phenol Red Lactose Sucrose Agar) is a highly selective and differential solid culture medium developed for the isolation and presumptive identification of Salmonella species (excluding S. Typhi and S. Paratyphi) from food, feed, and environmental samples.

Originally introduced by the German Federal Health Office (BGA), BPLS Agar is the official reference plating medium in historic §35 LMBG methods and is now incorporated into LFGB §64. It remains widely used across Europe as a confirmatory selective agar alongside XLD, HE, and Bismuth Sulfite Agar.

Selective & Differential Principle

Brilliant Green + Phenol Red
Strong inhibition of Gram-negative background flora and Proteus spp.

Lactose & Sucrose Fermentation

  • Fermenters → yellow colonies

  • Non-fermenting Salmonella → red to pink colonies

Hydrogen Sulfide Detection

  • H₂S-positive Salmonella strains form black centres, enhancing visual confirmation


Composition (per litre)

  • Peptone: 8.0 g

  • Meat Extract: 5.0 g

  • Yeast Extract: 3.0 g

  • Lactose: 10.0 g

  • Sucrose: 10.0 g

  • Sodium Chloride: 5.0 g

  • Phenol Red: 0.08 g

  • Brilliant Green: 0.0047 g (4.7 mg)

  • Agar: 12.0–15.0 g

Final pH: 6.9 ± 0.2 at 25 °C


Preparation

  1. Suspend 53.1–56.1 g of dehydrated medium in 1 L purified water

  2. Heat with frequent agitation and boil for 1 minute until dissolved

  3. Do not autoclave (brilliant green and phenol red are heat-labile)

  4. Cool to 45–50 °C, mix thoroughly

  5. Pour plates and allow to solidify


Expected Colony Appearance

(18–24 h at 35–37 °C)

  • Salmonella spp.: Pink-red to red colonies (2–4 mm), often with black centres

  • Coliforms / E. coli: Yellow colonies or inhibited

  • Proteus / Pseudomonas: Inhibited or very small pale colonies

  • Background flora: Strongly suppressed


Storage & Stability

  • Dehydrated medium: Store below 25 °C, dry, protected from light

  • Prepared plates: Store at 2–8 °C, protected from light; use within 7 days

For laboratory food and environmental microbiology use only.
Not intended for clinical or therapeutic applications.
