The ColiZist™ Kit (ACZ-1020) is a rapid, simultaneous detection system for total coliforms and Escherichia coli in water samples. Designed for routine microbiological water quality testing, the kit combines chromogenic and fluorogenic substrates in a single, easy-to-use format.
Each bottle is pre-measured for 100 mL water samples, allowing direct inoculation without media preparation. Principle of the Method
Detection Mechanism
Peptones and phosphate buffer support rapid coliform growth
Lauryl sulfate selectively suppresses Gram-positive bacteria
X-GAL (chromogenic substrate):
• Hydrolysed by coliform β-galactosidase → blue-green colour
MUG (fluorogenic substrate):
• Hydrolysed by E. coli β-glucuronidase → blue fluorescence under UV light

Test Procedure
General Method
Add the contents of one ColiZist™ bottle to 100 mL of water sample
Cap and mix gently until dissolved
Incubation Conditions
35–37 °C for 18–24 hours
or
20–25 °C for up to 48 hours
Interpretation of Results
Visual Assessment
Negative result:
No colour change (yellowish medium)
Total coliforms present:
Blue-green colour (X-GAL reaction)
E. coli present:
Blue-green colour with blue fluorescence under UV light (MUG reaction)
