Q1: Which bundle is right for a standard cloning project?
The Blue-White Screening & Cloning Kit (Core) covers the complete standard workflow — LB media for culture and plating, dual antibiotic selection (Ampicillin + Chloramphenicol), IPTG and X-Gal for blue-white screening, and base components for custom media preparation. It is the bundle most researchers reach for when starting a new cloning project.
Q2: Why is SOC Medium included in the Premium bundle but not the Core?
SOC Medium is specifically used for the post-transformation recovery step — the 37 °C incubation period immediately after heat shock or electroporation. It is not required for routine culture or plating. The Core bundle is designed for researchers who already have SOC or who use LB for recovery. The Premium bundle includes SOC for researchers who want to maximise transformation efficiency from the start.
Q3: What is the difference between LB Lennox and LB Miller formulations?
LB Lennox contains 5 g/L NaCl, while LB Miller contains 10 g/L NaCl. Both support excellent E. coli growth. Lennox is the standard choice for molecular cloning and transformation, where the lower osmolarity is marginally better tolerated by competent cells. Miller is often preferred for phage work and some expression systems. AuSaMicS bundles use the Lennox formulation by default — Miller versions (AS-1267, AS-1270) are available individually if required.
Q4: Can I use these bundles with E. coli strains other than DH5α?
Yes — all media and reagents are compatible with standard laboratory E. coli strains including DH5α, TOP10, DH10B, BL21(DE3), XL1-Blue, JM109, and most cloning and expression strains. Antibiotic concentrations should be adjusted per plasmid manufacturer recommendations (typically Ampicillin 50–100 µg/mL, Chloramphenicol 25–34 µg/mL, Kanamycin 25–50 µg/mL).
