LB Broth (Miller)
Luria-Bertani Broth, Miller Modification — General-Purpose Liquid Culture Medium
Catalogue No: AS-1270
E. coli & Molecular Biology Miller Formulation NaCl 10 g/L Liquid Culture Australian Made QC Certified
Overview

LB Broth (Miller), Luria-Bertani Broth Miller modification, is a nutrient-rich, general-purpose liquid culture medium widely used for the cultivation and maintenance of Escherichia coli in molecular biology and recombinant DNA applications. The Miller modification contains a higher sodium chloride concentration (10.0 g/L) supporting osmotic balance and membrane transport across standard E. coli strains.

The combination of tryptone and yeast extract supplies essential amino acids, peptides, vitamins, and growth factors, supporting robust growth including K-12 derivatives. LB Broth (Miller) is used as both a liquid medium and as a base for LB Agar (Miller) plate preparation.

Key Features
  • Standard Miller formulation — global default for routine E. coli liquid culture
  • Optimised NaCl (10.0 g/L) for osmotic balance and membrane transport
  • Supports rapid growth to high OD₆₀₀ for overnight and log-phase cultures
  • Suitable as liquid medium and as base for LB Agar (Miller) preparation
  • Easily modified with antibiotics, IPTG, sugars, or other supplements
  • Compatible with all standard E. coli K-12 and B strains
Applications
  • Routine Liquid Culture: Overnight cultures for miniprep, midiprep, maxiprep; mid-log phase for transformation
  • Competent Cell Preparation: Chemical (CaCl₂) and electrocompetent E. coli preparation
  • Protein Expression: Pre-induction liquid culture for BL21, Rosetta, ArcticExpress
  • Plasmid Maintenance: Propagation and amplification of plasmid-bearing strains
  • Agar Base: Add 15.0 g/L agar before autoclaving to prepare LB Agar (Miller)
  • Teaching: Standard molecular biology liquid culture instruction
Preparation
  1. Dissolve 25.0 g in 1 litre of demineralised or purified water.
  2. Mix or heat gently with agitation until fully dissolved.
  3. Adjust pH to 7.0 ± 0.2 if required.
  4. Sterilise by autoclaving at 121 °C for 15 minutes.
  5. Cool to room temperature before use or inoculation.
  6. Add filter-sterilised antibiotics or supplements aseptically as required.

For LB Agar (Miller): Add 15.0 g/L agar before autoclaving. Cool to 45–50 °C, add antibiotics, pour 20–25 mL per 90 mm plate.
