Product Description

Orange Serum Broth (OSB) is a selective liquid culture medium developed for the isolation, cultivation, and enumeration of aciduric microorganisms associated with the spoilage of fruit juices, soft drinks, wines, ciders, and other acidic food and beverage products.

The formulation supports the growth of acid-tolerant bacteria, yeasts, and moulds, including Lactobacillus, Leuconostoc, Acetobacter, and Gluconobacter species. The medium is widely used in beverage production facilities, wineries, breweries, juice processing plants, and contract microbiology laboratories for routine quality control testing.

Principle of the Medium

Functional Components

Casein Enzymic Hydrolysate (Peptone): Provides nitrogen, peptides, and amino acids
Yeast Extract: Supplies vitamins and growth factors
Dextrose: Readily fermentable carbohydrate source
Dipotassium Hydrogen Phosphate: Buffering component
Orange Serum Solids: Provide natural organic acids and carbon sources characteristic of citrus products

The low final pH (approximately 5.5) suppresses most non-aciduric background flora while enabling robust growth of targeted spoilage microorganisms.

Preparation Instructions

Medium Preparation

Suspend the required quantity of dehydrated medium in purified water
Heat with agitation until fully dissolved
Sterilise by autoclaving at 121 °C for 15 minutes
Cool to room temperature before use
