Overview

Tryptic Soy Agar with Lecithin and Polysorbate 20 (AS-1434) is the solid plating counterpart of SCDLP20/TAT Broth. It is a TSB-based agar supplemented with Lecithin (0.7 g/L) and Polysorbate 20 (Tween 20, 5 mL/L) as neutralising agents. It is used for colony isolation, plate counting, and environmental surface monitoring in cosmetics and pharmaceutical QC workflows, particularly after SCDLP20 Broth pre-enrichment.

Mode of Action
Dual Neutralisation on Solid Medium → Colony Recovery & Enumeration

Soya Lecithin (0.7 g/L) neutralises cationic QACs, cetrimide, chlorhexidine, and polymyxin B. Polysorbate 20 (5 mL/L) neutralises phenolics, parabens, benzoic acid esters, and in combination with lecithin also ethanol. At these concentrations, both agents are effective without inhibiting colony formation. The TSB nutritional base (casein + soy peptone) provides the amino acids, vitamins, and growth factors for broad microbial recovery on the agar surface.

Preparation Protocol
1 Suspend approximately 41 g of dehydrated base in 950 mL distilled water. Heat with stirring until completely dissolved.
2 Add 5 mL Polysorbate 20 (Tween 20). Mix until homogeneous. Adjust to 1 litre final volume.
3 Adjust pH to 7.3 ± 0.2 if required.
4 Autoclave at 121°C for 15 minutes. Do not over-autoclave — excessive heat reduces agar gel strength.
5 Cool to 45–50°C in a water bath. Pour ~20 mL per 90 mm Petri dish on a level surface. Allow to solidify.
6 Dry plates at 37°C / 30 min (lids ajar). Store at 2–8°C in sealed bags. Discard plates showing contamination, desiccation, or cracking.
⚠️ Slight opalescence of prepared plates is normal due to lecithin — does not affect performance. Do not incubate plates directly from 2–8°C storage; equilibrate to room temperature to prevent condensation on colonies.
