2× YT Broth is a nutrient-rich, non-selective liquid culture medium formulated to support rapid growth and high cell density of bacteria, particularly Escherichia coli used in molecular biology and recombinant DNA applications. Compared with standard LB Broth, 2× YT contains double the tryptone (16.0 g/L) and yeast extract (10.0 g/L), promoting faster growth rates, higher biomass yield, and improved plasmid production.

Preparation
- Dissolve 31.0 g of dehydrated medium in 1 litre of purified water with agitation.
- Heat until completely dissolved. Adjust pH to 7.0 ± 0.2 if required.
- Dispense into suitable culture flasks or bottles.
- Sterilise by autoclaving at 121 °C for 15 minutes.
- Cool to room temperature before inoculation.
- Add filter-sterilised antibiotics aseptically as required.
