Q2: Which antibiotic should I choose?
Selection depends on the resistance marker carried by your vector or plasmid — not on the cloning workflow itself. Check your vector map or plasmid documentation and use the selection conditions specified by the applicable protocol.
Q3: Do I need SOC Medium?
SOC Medium is used for the post-transformation recovery step — the incubation period immediately after heat shock or electroporation, before plating. It's not required for every protocol; some labs use LB for recovery instead. Add it if your transformation protocol calls for a dedicated rich recovery medium.
Q4: What are IPTG and X-Gal used for?
IPTG is a non-metabolisable inducer of lac-controlled expression, while X-Gal is a chromogenic β-galactosidase substrate. Together they enable blue-white colony screening in a compatible host/vector system with an appropriate lacZ α-complementation system. Confirm that your strain and vector support this before relying on colony colour for screening.
Q5: What's the difference between LB Lennox and LB Miller formulations?
LB Lennox contains 5 g/L NaCl, while LB Miller contains 10 g/L NaCl. Both support E. coli growth; Lennox is the formulation used in this set by default. Miller versions (AS-1267, AS-1270) are available individually if your protocol specifies it.
