Overview

A1 Medium, also known as ATCC Medium No. 1, is a highly enriched, non-selective liquid culture medium formulated for the maximum recovery and luxuriant growth of fastidious and delicate microorganisms. It is particularly effective for specimens containing low bacterial counts or stressed, injured, or antibiotic-exposed cells.

This medium is widely used in clinical and research microbiology laboratories for the enrichment and primary recovery of demanding bacterial species, including Streptococcus, Neisseria, Haemophilus, and Brucella. Its complex formulation supplies essential amino acids, vitamins, sulphur compounds, and growth factors, ensuring reliable growth even under challenging conditions.

Principle of the Medium

A1 Medium provides a rich blend of peptones, yeast extract, carbohydrates, and sulphur-containing compounds that support metabolic recovery and rapid multiplication of nutritionally demanding bacteria. The inclusion of reducing agents aids in the recovery of oxygen-sensitive or injured cells, making the medium especially effective for primary isolation.


Composition (per litre)

  • Trypticase Peptone (Pancreatic Digest of Casein) – 20.0 g

  • Yeast Extract – 5.0 g

  • Glucose – 5.0 g

  • Sodium Chloride (NaCl) – 5.0 g

  • L-Cystine – 0.2 g

  • Sodium Sulphite (Na₂SO₃) – 0.1 g

  • Thiouracil – 0.01 g

  • Phenol Red (optional indicator) – 0.01 g

Final pH: 7.2 ± 0.2 at 25 °C


Preparation

  1. Suspend 35.31 g of dehydrated medium in 1 L purified water

  2. Mix thoroughly and warm gently until fully dissolved

  3. Dispense into tubes or bottles as required

  4. Sterilize by autoclaving at 121 °C for 15 minutes

  5. Cool to room temperature before use


Storage & Stability

  • Dehydrated medium: Store below 30 °C in a cool, dry place, protected from light

  • Prepared broth: Store at 2–8 °C and use within 14 days
