Overview
Alkaline Peptone Water (APW) is a selective enrichment broth formulated for the isolation and recovery of Vibrio species, particularly Vibrio cholerae and Vibrio parahaemolyticus, from clinical specimens, seafood, and environmental water samples.
The medium’s high alkaline pH (8.4–8.6), combined with peptone and sodium chloride, promotes rapid multiplication of Vibrio spp. while inhibiting most competing enteric bacteria, including Enterobacteriaceae. APW is internationally recognised as the primary enrichment medium recommended by WHO, CDC, FDA, and ISO 21872-1 prior to plating on selective agars such as TCBS.

Principle of the Medium
Vibrio species exhibit rapid growth under alkaline conditions and tolerate elevated salt concentrations. APW exploits these characteristics by combining a nutrient-rich peptone base with alkaline pH, suppressing competing intestinal flora while allowing Vibrio spp. to proliferate before selective plating.
Composition (per litre)

Peptone (Bacteriological Peptone) – 10.0 g

Sodium Chloride (NaCl) – 10.0 g
Final pH: 8.5 ± 0.2 at 25 °C
(Adjusted with 1 N NaOH if required)
Preparation
-
Dissolve 10 g peptone and 10 g NaCl in 1 L distilled or deionised water

Mix thoroughly until completely dissolved

Adjust pH to 8.5 using 1 N NaOH if necessary

Dispense into tubes or flasks (commonly 10 mL volumes)

Sterilise by autoclaving at 121 °C for 15 minutes

Cool to room temperature before use
Common Variants

Marine / estuarine Vibrio (e.g. V. parahaemolyticus):
NaCl increased to 20–30 g/L -
Less halophilic Vibrio (e.g. V. vulnificus):
NaCl reduced to 5 g/L
Incubation

35–37 °C for 6–18 hours (primary enrichment)

A second enrichment (subculture into fresh APW) is recommended for heavily contaminated samples
