Mode of Action

Fenbendazole exerts its primary pharmacological action through selective binding to β-tubulin, the structural subunit of microtubules. This binding inhibits tubulin polymerisation, disrupting the cytoskeletal network and mitotic spindle apparatus. In helminth models, this impairs glucose uptake, energy metabolism, and cell division.

Fenbendazole binds β-tubulin (colchicine-binding site)

Inhibition of tubulin polymerisation

Disruption of mitotic spindle & cytoskeletal integrity

Impaired glucose transport & energy depletion

Cell cycle arrest / cell death (parasite or target cell)

Fenbendazole binds with higher selectivity to parasite tubulin than to mammalian tubulin isoforms — making it a valuable tool for species-specific tubulin pharmacology research. At research concentrations it is routinely used as a positive control for microtubule disruption assays.

Technical Overview
Pharmacological Class Benzimidazole carbamate; β-tubulin polymerisation inhibitor
Primary Target β-tubulin (colchicine-binding site)
IC₅₀ (tubulin polymerisation) ~1–10 µM (cell-line and species dependent)
Selectivity Parasite > mammalian tubulin (structure-dependent)
Metabolites of Interest Fenbendazole sulfoxide (oxfendazole); fenbendazole sulfone
Typical Working Conc. 0.1–50 µM depending on assay system
Vehicle Recommendation Dissolve in DMSO to 10–30 mM stock; dilute in buffer/media (<0.1% DMSO final)
