DCLS Agar (Desoxycholate Citrate Lactose Sucrose Agar) | Selective Medium for Enteric Pathogens | AS‑1192
DCLS Agar (Desoxycholate Citrate Lactose Sucrose Agar)
Selective & Differential Medium for Isolation of Enteric Pathogens (Salmonella & Shigella)
DCLS Agar is a selective and differential medium designed for the isolation of enteric pathogens, particularly Salmonella and Shigella species, from clinical, food, and environmental samples. The formulation combines desoxycholate and citrate salts for inhibition with lactose and sucrose for differentiation of fermenting organisms.
🏆 Enteric Pathogen Medium
🔬 Principle of the Medium
DCLS Agar contains sodium desoxycholate and citrate salts which inhibit Gram-positive bacteria and many non-enteric organisms, allowing selective recovery of enteric pathogens.
Lactose and sucrose serve as fermentable carbohydrates, enabling differentiation based on acid production. Fermenters produce colored colonies, while non-fermenters such as Salmonella and Shigella remain pale or colorless.

Non-fermenters → Pale colonies (pathogens)
🧪 Applications
- Isolation of Salmonella spp. and Shigella spp.
- Food safety testing (meat, dairy, processed foods)
- Clinical stool sample analysis
- Environmental and water microbiology
📊 Enteric Media Comparison Guide
| Medium | Selectivity | Differentiation | Best Use |
|---|---|---|---|
| DCLS Agar | High | Lactose + sucrose | Classical enteric isolation |
| XLD Agar | High | H2S + xylose | Modern Salmonella detection |
| Hektoen Enteric Agar | Moderate–High | Multiple sugars + H2S | Clinical stool screening |
| MacConkey Agar | Moderate | Lactose | General enterics |
💡 Practical Notes
- Highly selective – may suppress stressed organisms
- Use with enrichment broth for best recovery
- Clear differentiation between fermenters and pathogens
- Classical alternative to XLD and HE agar
