Overview

LB Agar (Miller) is the most widely used general-purpose solid culture medium for the routine cultivation, cloning, and maintenance of recombinant Escherichia coli strains in molecular biology laboratories. The Miller formulation (1972) contains 10.0 g/L sodium chloride — the global standard for routine E. coli work.

Fully compatible with common antibiotics, IPTG/X-gal reporter systems, and all standard laboratory E. coli strains. Supports rapid bacterial growth, high cell density, and reliable plasmid maintenance.

Applications
  • Routine Plating: DH5α, BL21, TOP10, JM109, XL1-Blue and other standard E. coli strains
  • Antibiotic Selection: Ampicillin, kanamycin, chloramphenicol, tetracycline, gentamicin selection plates
  • Blue–White Screening: X-gal (40 µg/mL) + IPTG (0.1 mM) for recombinant colony identification
  • Plasmid Propagation: Colony isolation, transformation recovery plating, and overnight culture preparation
  • Phage Work: Bacteriophage propagation, plaque assay overlay plates
  • Teaching: Standard molecular biology instruction and microbiology cultivation
Preparation
  1. Suspend 40.0 g in 1 litre of demineralised or purified water.
  2. Heat with agitation until fully dissolved.
  3. Adjust pH to 7.0 ± 0.2 if required (1 M NaOH or 1 M HCl).
  4. Sterilise by autoclaving at 121 °C for 15 minutes.
  5. Cool to 45–50 °C.
  6. Add filter-sterilised antibiotics or supplements aseptically.
  7. Dispense 20–25 mL per 90 mm sterile Petri dish.
  8. Allow to solidify on a level surface before use.
