Description

MacConkey Agar with Sorbitol (AS-1284) is a selective and differential dehydrated culture medium primarily used for the isolation and identification of Enterobacteriaceae, including pathogenic Escherichia coli O157:H7. Sorbitol replaces lactose as the fermentable carbohydrate, enabling differentiation of sorbitol-fermenting from non-sorbitol-fermenting bacteria.

Non-sorbitol fermenters such as E. coli O157:H7 produce colourless, translucent colonies, while sorbitol-fermenting coliforms produce pink to red colonies due to acid production from sorbitol fermentation and the resulting colour change of the neutral red pH indicator.

Preparation Instructions
SuspensionSuspend 48.5 g in 1 litre of purified water. Mix thoroughly. HeatingHeat with agitation and boil for 1 minute until dissolved. Do NOT autoclave. DispensingCool to 45–50 °C. Pour ~20 mL per sterile Petri dish. Incubation35–37 °C, aerobic, 18–24 hours. Yield100 g → ~100 plates  |  500 g → ~500 plates
