Sabouraud Dextrose Agar with Chloramphenicol

Selective Fungal Culture Medium for Yeasts & Molds
Catalog Number: AS-1342

Overview

Sabouraud Dextrose Agar with Chloramphenicol (SDA + CAP) is a selective solid culture medium formulated for the isolation, cultivation, and enumeration of yeasts and molds from specimens with high bacterial background flora.

The medium combines an acidic pH (~5.6) and high dextrose concentration to promote robust fungal growth, while chloramphenicol provides broad-spectrum inhibition of Gram-positive and Gram-negative bacteria. This makes SDA + Chloramphenicol particularly effective for mixed or contaminated samples encountered in clinical, pharmaceutical, cosmetic, food, and environmental microbiology.

Key Features & Benefits

  • Acidic pH (~5.6) suppresses most bacterial contaminants

  • High dextrose content enhances fungal metabolism and recovery

  • Chloramphenicol provides broad-spectrum antibacterial inhibition

  • Excellent performance with contaminated or mixed specimens

  • Well-established, internationally used selective fungal medium

Typical Applications

Clinical Mycology

  • Isolation of Candida, Aspergillus, dermatophytes, and other pathogenic fungi

Pharmaceutical Quality Control

  • Microbial Limit Tests (USP / EP / JP compliant) for non-sterile products

Cosmetic & Personal Care Microbiology

  • Fungal enumeration and stability testing

Food & Beverage Microbiology

  • Detection and enumeration of yeasts and molds in products with bacterial background

Environmental Microbiology

  • Recovery of fungi from air, surface, and environmental samples

Principle of the Medium

Sabouraud Dextrose Agar provides a high-carbohydrate, low-pH environment that favours fungal growth while inhibiting bacteria. The addition of chloramphenicol further suppresses bacterial contaminants, allowing reliable recovery and enumeration of yeasts and molds even from heavily contaminated samples.

Composition (per litre)

  • Peptones (casein / animal origin): 10.0 g

  • Dextrose: 40.0 g

  • Agar: 15.0 g

  • Chloramphenicol: 0.05–0.1 g (0.005–0.01%)

Final pH: 5.6 ± 0.2 at 25 °C

Preparation

  1. Suspend 65 g of dehydrated medium in 1 L purified water

  2. Heat with agitation and boil until completely dissolved

  3. Sterilise by autoclaving at 121 °C for 15 minutes

  4. Cool to 45–50 °C

  5. If chloramphenicol is supplied separately, aseptically add the sterile solution and mix well

  6. Pour into sterile Petri dishes or tubes and allow to solidify

Incubation

  • Yeasts and molds: 25–30 °C for 3–7 days

  • Dermatophytes: up to 14–21 days

Interpretation

  • Yeasts: Smooth, creamy, moist colonies

  • Molds: Filamentous, cottony, or granular colonies

  • Bacteria: Growth inhibited by chloramphenicol

Storage & Stability

  • Dehydrated medium: Store 15–30 °C, dry and tightly sealed

  • Prepared plates: Store at 2–8 °C, protected from light; usable for up to 3 months

Intended Use

For laboratory use only, including clinical, pharmaceutical, cosmetic, food, and environmental microbiology.
Not for human or veterinary use. Not for consumption.

Quality & Compliance

  • Manufactured under controlled conditions for consistent performance

  • Suitable for USP, EP, and JP microbial limit testing

  • Performance comparable to leading international brands

  • COA and SDS available upon request
