Overview
Sabouraud Dextrose Agar with Chloramphenicol (SDA + CAP) is a selective solid culture medium formulated for the isolation, cultivation, and enumeration of yeasts and molds from specimens with high bacterial background flora.
The medium combines an acidic pH (~5.6) and high dextrose concentration to promote robust fungal growth, while chloramphenicol provides broad-spectrum inhibition of Gram-positive and Gram-negative bacteria. This makes SDA + Chloramphenicol particularly effective for mixed or contaminated samples encountered in clinical, pharmaceutical, cosmetic, food, and environmental microbiology.

Principle of the Medium
Sabouraud Dextrose Agar provides a high-carbohydrate, low-pH environment that favours fungal growth while inhibiting bacteria. The addition of chloramphenicol further suppresses bacterial contaminants, allowing reliable recovery and enumeration of yeasts and molds even from heavily contaminated samples.
Composition (per litre)

Peptones (casein / animal origin): 10.0 g

Dextrose: 40.0 g

Agar: 15.0 g

Chloramphenicol: 0.05–0.1 g (0.005–0.01%)
Final pH: 5.6 ± 0.2 at 25 °C
Preparation
-
Suspend 65 g of dehydrated medium in 1 L purified water

Heat with agitation and boil until completely dissolved

Sterilise by autoclaving at 121 °C for 15 minutes

Cool to 45–50 °C

If chloramphenicol is supplied separately, aseptically add the sterile solution and mix well

Pour into sterile Petri dishes or tubes and allow to solidify
Incubation

Yeasts and molds: 25–30 °C for 3–7 days

Dermatophytes: up to 14–21 days
Interpretation
-
Yeasts: Smooth, creamy, moist colonies

Molds: Filamentous, cottony, or granular colonies

Bacteria: Growth inhibited by chloramphenicol
Storage & Stability

Dehydrated medium: Store 15–30 °C, dry and tightly sealed

Prepared plates: Store at 2–8 °C, protected from light; usable for up to 3 months
