TBX Agar is a selective chromogenic medium used for the detection and enumeration of Escherichia coli based on β-glucuronidase (GUD) activity. E. coli colonies produce a characteristic blue-green coloration due to hydrolysis of the chromogenic substrate X-glucuronide, enabling rapid and reliable identification.

🔬 Principle of the Medium

TBX Agar contains tryptone as a nutrient source and bile salts to inhibit Gram-positive organisms. The key component is X-glucuronide, a chromogenic substrate that is cleaved by β-glucuronidase produced by most E. coli strains.

Upon hydrolysis, an insoluble chromophore is released, producing blue-green colonies, allowing direct identification without additional confirmatory tests.
