Description

Tryptone Sulfite Neomycin (TSN) Agar is a specialized selective and differential medium engineered for the primary isolation and enumeration of Clostridium perfringens. This refined formulation incorporates dual-antibiotic selectivity and an integrated iron indicator system, making it a critical tool for food safety, dairy microbiology, and water quality laboratories.

Target Mechanism
H₂S Differentiation
Sodium Sulfite and Ammonium Ferric Citrate react to form black precipitates around sulfite-reducing colonies, allowing for visual confirmation of Clostridia.
Dual Selectivity
A precise blend of Neomycin and Polymyxin B suppresses a wide spectrum of competing Gram-positive and Gram-negative flora.
Nutrient Base
Tryptone and Yeast Extract provide high concentrations of essential amino acids and B-complex vitamins to support rapid anaerobic development.

Preparation Procedure
Reconstitution Suspend 41.07 g of medium in 1 L of purified water.
Dissolution Heat with gentle agitation until completely dissolved.
Sterilization Autoclave at 121°C for 15 minutes.
Pouring Cool to 45–50°C before dispensing into sterile Petri dishes.
Incubation & Interpretation

Optimal results require anaerobic incubation at 37°C for 18–24 hours. Some strains may require up to 48 hours for complete H₂S reaction.
