Overview

Arginine Brilliant-Green Glucose Peptone Broth (ABGP) is a selective enrichment medium originally developed by Schubert (1966) for the isolation of Streptococcus agalactiae (Group B Streptococcus, GBS) from mixed microbial populations. It has been used for enrichment from clinical specimens (vaginal and rectal swabs, neonatal samples) as well as raw milk and dairy products.

The selectivity of ABGP Broth is achieved through the combined action of arginine, brilliant-green dye, and nalidixic acid, which strongly inhibit Gram-negative bacteria, enterococci, staphylococci, and most non-GBS streptococci. Under these conditions, GBS grows luxuriantly and produces a characteristic colour change from purple to yellow, resulting from arginine deamination followed by glucose fermentation.

ABGP Broth has been recommended in several older European protocols (notably in Germany, Switzerland, and Austria) as an alternative or complementary enrichment medium to LIM or Todd–Hewitt broth.

Composition (per litre)

  • Peptone – 10.0 g

  • Glucose – 5.0 g

  • L-Arginine hydrochloride – 5.0 g

  • Sodium chloride – 5.0 g

  • Disodium hydrogen phosphate (Na₂HPO₄) – 2.0 g

  • Brilliant Green – 0.01 g (10 mg)

  • Nalidixic Acid – 0.015 g (15 mg)

  • Bromocresol Purple – 0.02 g

Final pH: 7.2 ± 0.2 at 25 °C


Preparation

  • Dissolve all components except nalidixic acid and bromocresol purple in 900 mL distilled water

  • Adjust pH to 7.2

  • Add bromocresol purple dissolved in a few drops of 0.1 N NaOH

  • Make up to 1 L with distilled water

  • Dispense 5–10 mL into tubes

  • Sterilise by autoclaving at 121 °C for 15 minutes

  • After cooling to below 60 °C, aseptically add filter-sterilised nalidixic acid (15 mg/L)

(Some commercial formulations may include heat-stable nalidixic acid derivatives.)


Incubation & Interpretation

  • Incubation: 35–37 °C for 18–24 hours (up to 48 hours if required)

  • Positive reaction: Medium turns yellow (presumptive GBS growth)

  • Negative reaction: Medium remains purple


Subculture & Confirmation

Presumptive positive cultures should be subcultured onto:

  • Columbia CNA blood agar

  • Granada medium

  • CHROMagar™ StrepB


Storage & Stability

  • Prepared broth (with nalidixic acid): Store at 2–8 °C, protected from light

  • Shelf life: Use within 4 weeks
