Overview
AC Broth (Actinomycete Isolation Broth) is a selective liquid culture medium formulated for the enrichment and cultivation of actinomycetes, including Streptomyces, Nocardia, Actinomadura, and other rare actinobacterial genera.
It is the liquid counterpart of Actinomycete Isolation Agar (AC Agar) and is specifically designed for use with heavily contaminated environmental samples such as soil, plant material, sediments, and compost. The formulation suppresses competing Gram-negative bacteria, many Gram-positive bacteria, and fast-growing fungi, allowing slow-growing actinomycetes to proliferate during extended incubation.
AC Broth is widely used as a pre-enrichment medium prior to plating on selective solid media and in antibiotic discovery and actinomycete ecology programs.

Principle of the Medium
AC Broth utilises selective nitrogen sources (sodium caseinate and L-asparagine) and sodium propionate to inhibit competing microorganisms. An alkaline pH and optional antifungal agents further enhance selectivity. Glycerol, supplied separately, serves as a preferred carbon source for many actinomycetes, supporting their enrichment in liquid culture.

Magnesium Sulfate (MgSO₄·7H₂O) – 0.1 g

Ferrous Sulfate (FeSO₄·7H₂O) – 0.001 g
Final pH: 8.0–8.2 at 25 °C

Glycerol – 5.0 g (≈ 5 mL)

Cycloheximide (Actidione) – 50 mg (filter-sterilised)

Nystatin (optional, for heavy fungal contamination) – 50 mg (filter-sterilised)
Preparation
-
Suspend 11.7 g of basal medium in 1 L distilled or deionised water

Adjust pH to 8.1 with 1 N NaOH if required

Sterilise by autoclaving at 121 °C for 15 minutes

Cool to 45–50 °C

Aseptically add filter-sterilised glycerol and antibiotic solutions

Dispense into sterile tubes or flasks
Incubation

Temperature: 25–30 °C

Agitation: 100–150 rpm

Duration: 5–14 days

Extended incubation (up to 4 weeks) may be required for rare or slow-growing genera
Storage & Shelf Life

Dehydrated medium: Store below 30 °C, dry and protected from light

Prepared broth (with antibiotics): Store at 2–8 °C and use within 2 weeks
