Preparation Protocol
- 1 Suspend 36.0–38.0 g of BCYE base in 950 mL purified water.
- 2 Add ACES buffer and α-ketoglutarate; mix thoroughly.
- 3 Adjust pH to 6.9 at 50 °C using 1 N KOH.
- 4 Autoclave at 121 °C for 15 minutes.
- 5 Cool to 50 °C, then aseptically add filter-sterilized L-cysteine and iron solutions.
- 6 Mix well and pour plates.
