Overview

BG11 Broth is a chemically defined liquid culture medium universally accepted for the axenic cultivation of freshwater cyanobacteria and selected unicellular green algae. Originally formulated by Stanier and colleagues and subsequently standardised by Rippka et al., BG11 remains the gold-standard reference medium in phycology, biotechnology, and aquatic ecology.

The medium is optimised for photoautotrophic growth, using nitrate as the sole nitrogen source, no combined organic carbon, and carefully balanced trace metals. It is widely used for cultivation of genera such as Synechococcus, Anabaena, Nostoc, Microcystis, and Spirulina, as well as for cyanobacterial genomics, physiology, and toxicity testing.

Principle of the Medium

BG11 Broth provides all essential inorganic nutrients required for cyanobacterial growth while excluding organic carbon sources. Growth is driven by photosynthesis, with nitrate supplying nitrogen and a low-phosphate background minimising precipitation and metabolic interference. A defined trace-metal system (A5) supports enzymatic and photosynthetic processes while maintaining clarity for optical measurements.

Directions for Use

Suspend 1.6894 g of dehydrated medium per litre of purified or distilled water.

Heat if necessary to fully dissolve the medium.

Adjust pH with 1 M NaOH or HCl if it does not achieve 7.1.

Dispense in flasks or as desired.

Sterilise by autoclaving at 121°C for 15 minutes.

Cool the medium to room temperature before use.
Marine Species Supplementation
For marine species, prepare a solution of 10 g/L sodium chloride and 1 g/L Vitamin B12. Add 20 mL of this solution (sterile filtered) to 1000 mL of the prepared BG11 medium.
Incubation Conditions
Light Intensity 2,000–3,000 lux (neon light source is sufficient)
Photoperiod Continuous illumination (24 hours/day) optimal for maintenance
Temperature 20–25°C (room temperature)
Vessel Covering Flasks may be covered with grease-proof paper during incubation
Storage & Stability
Prepared medium: store at 2–8°C, protected from light. Use within 3 months under recommended conditions.
