Bifidobacterium Broth provides a reduced, nutrient-rich environment suitable for anaerobic bifidobacteria:

  • Tryptone and peptone supply peptides, amino acids, and nitrogen

  • Yeast extract provides vitamins, nucleotides, and trace nutrients

  • Tomato juice solids contribute organic acids and additional growth factors

  • Glucose serves as a readily fermentable energy source

  • Tween 80 enhances nutrient uptake and cell membrane function

Together, these components support consistent growth and maintenance of bifidobacteria under anaerobic conditions.


Composition (Typical – per litre)

  • Tryptone: 20.0 g

  • Peptone: 10.0 g

  • Yeast extract: 10.0 g

  • Tomato juice solids: 16.65 g

  • Dextrose (glucose): 20.0 g

  • Polysorbate 80 (Tween 80): 2.0 g

Optional (for solid medium):

  • Agar: 15.0 g

Final pH at 25 °C: 6.8 ± 0.2


Preparation (Typical)

  • Suspend 78.65 g of dehydrated medium in 1 L purified or distilled water

  • Heat gently if required to dissolve completely

  • Dispense into tubes or flasks

  • Sterilise by autoclaving at 121 °C for 15 minutes

  • Cool and use under anaerobic conditions


Incubation

  • Atmosphere: Anaerobic

  • Temperature: 35–37 °C

  • Incubation time: 24–72 hours, depending on strain and application


Storage & Stability

Dehydrated medium

  • Store below 30 °C in a tightly closed, dry container

Prepared broth

  • Store at 2–8 °C

  • Use before the stated expiry date
