Skirrow’s Campylobacter Agar (AS-1165) | Clinical Recovery Medium
Skirrow's Campylobacter Agar
Selective Blood-Enriched Medium for Primary Isolation of Campylobacter spp.
A specialized blood-enriched selective agar for the primary recovery of Campylobacter jejuni, C. coli, and related thermotolerant campylobacters from specimens with heavy background flora. Built around the classic VPT selective system with blood enrichment for dependable clinical and food microbiology workflows.
Selectivity Specialist
Overview
Skirrow's Campylobacter Agar is a classical selective blood-containing medium developed for the primary isolation of thermotolerant Campylobacter species from mixed specimens, especially feces and other enteric samples. The medium is designed to suppress much of the competing intestinal flora while still permitting recovery of delicate microaerophilic campylobacters.
Its performance depends on a nutritive agar base enriched with blood and supplemented with the traditional VPT antibiotic system: vancomycin, polymyxin B, and trimethoprim. When combined with incubation at approximately 42°C under a microaerophilic atmosphere, the medium creates a selective ecological niche that favors thermotolerant campylobacters over many enteric competitors.
Skirrow medium remains valuable in clinical and food microbiology where a classical blood-enriched Campylobacter medium is preferred, especially in workflows prioritizing recovery of stressed organisms from heavily contaminated samples.
Applications
- Primary isolation of Campylobacter jejuni and C. coli from stool samples
- Recovery of thermotolerant Campylobacter from clinical enteric specimens
- Use in food microbiology and poultry-related Campylobacter screening workflows
- Selective culture where heavy enteric commensal flora must be suppressed
- Routine and reference laboratory Campylobacter culture programs
Medium Formulation & Selective Dynamics
Base Composition (per Liter)
| Ingredient | Concentration | Function | Technical Role |
|---|---|---|---|
| Peptone Special | 23.0 g | Nutrient base | Provides amino acids and nitrogen for Campylobacter growth |
| Sodium Chloride | 5.0 g | Osmotic balance | Maintains physiological stability |
| Corn Starch | 1.0 g | Detoxification support | Can absorb toxic metabolites and support recovery |
| Agar No. 2 | 12.0 g | Gelling matrix | Provides firm selective plating surface |
Complete Medium Requirements
The dehydrated base alone is not the final working medium. For performance consistent with classical Skirrow methodology, the prepared base is typically supplemented with:
- 5–7% lysed or defibrinated blood for enrichment and recovery support
- Selective supplement (VPT) containing vancomycin, polymyxin B, and trimethoprim
Mechanism of Selectivity
Skirrow's Campylobacter Agar operates through a triple-antibiotic selective system designed to suppress competing flora while preserving the viability of microaerophilic Campylobacter species:
- Vancomycin: inhibits many Gram-positive organisms
- Polymyxin B: suppresses a range of Gram-negative enteric competitors
- Trimethoprim: helps prevent swarming by Proteus species and reduces nuisance overgrowth
Combined with blood enrichment and incubation at 42°C in a microaerophilic atmosphere, the medium creates a strongly favorable growth niche for thermotolerant campylobacters.
Application Performance Matrix
| Organism | Expected Growth | Typical Colonial Morphology | Interpretation |
|---|---|---|---|
| C. jejuni ATCC 33291 | Excellent | Grey-white, flat, moist, spreading | Typical recovery target |
| C. coli ATCC 33559 | Excellent | Creamy, convex to glistening | Expected thermotolerant Campylobacter growth |
| Escherichia coli | Marked inhibition | None to minimal | Suppressed by selectives |
| Staphylococcus aureus | Complete to strong inhibition | No significant growth | Gram-positive suppression expected |
Campylobacter Media Comparison
| Medium | Selectivity | Key Strength | Limitation | Best Use |
|---|---|---|---|---|
| Skirrow Agar | High (VPT + blood) | Excellent recovery from stool and heavy-flora samples | Requires blood and selective supplement | Classical clinical Campylobacter isolation |
| CCDA Agar | Very high | Widely used in standardized food methods | Some stressed cells may recover less efficiently | Routine food testing / ISO workflows |
| Butzler Agar | Moderate | Established classical selective medium | Can be less selective in heavy-flora specimens | Traditional Campylobacter recovery |
Selection Guide
• Clinical stool samples carry heavy competing flora
• Blood-enriched recovery is preferred
• A classical validated Campylobacter workflow is desired
• ISO-aligned food methods are primary
• Blood-free selective workflow is preferred
• High-throughput routine screening is needed
• Campylobacter is not the main target
• A general enteric medium is needed
• Supplement management is impractical
