GN Broth (Gram-Negative Broth) | Selective Enrichment Media | Ausamics
Gram Negative Broth, GN Broth
AuSaMicS Life Science™ | www.ausamics.com.au
GN Broth is a selective enrichment medium used for the isolation and recovery of Shigella and Salmonella from a wide variety of sample types. Peptone serves as the principal source of nutrients, while sodium deoxycholate and sodium citrate suppress the growth of many Gram-positive organisms and certain competing Gram-negative bacteria.
Phosphate buffering helps maintain pH stability during incubation, supporting consistent enrichment performance. Recovery of target pathogens is enhanced when mannitol is used at a higher concentration instead of dextrose, as this helps inhibit Proteus growth. For improved isolation and confirmation, GN Broth is commonly used together with selective and non-selective agar media.
- Selective enrichment of enteric pathogens from clinical specimens
- Support medium for isolation workflows targeting Salmonella and Shigella
- Recovery of target pathogens from food and environmental monitoring samples
- Useful in routine quality control and contamination investigations
- Selective enrichment of Gram-negative enteric bacteria from water samples
- Applicable in hygiene, sanitation, and microbiological surveillance programs
| Step | Procedure |
|---|---|
| 1 | Suspend the recommended quantity of dehydrated medium in purified water according to the approved formulation or label instructions. |
| 2 | Heat with gentle agitation until the medium is completely dissolved. |
| 3 | Dispense into suitable sterile containers or tubes for enrichment use. |
| 4 | Sterilize using validated laboratory procedures appropriate for the medium formulation. |
| 5 | Cool to the recommended inoculation temperature before use. |
