Description

GN Broth is a selective enrichment medium used for the isolation and recovery of Shigella and Salmonella from a wide variety of sample types. Peptone serves as the principal source of nutrients, while sodium deoxycholate and sodium citrate suppress the growth of many Gram-positive organisms and certain competing Gram-negative bacteria.

Phosphate buffering helps maintain pH stability during incubation, supporting consistent enrichment performance. Recovery of target pathogens is enhanced when mannitol is used at a higher concentration instead of dextrose, as this helps inhibit Proteus growth. For improved isolation and confirmation, GN Broth is commonly used together with selective and non-selective agar media.

Preparation
Step Procedure
1 Suspend the recommended quantity of dehydrated medium in purified water according to the approved formulation or label instructions.
2 Heat with gentle agitation until the medium is completely dissolved.
3 Dispense into suitable sterile containers or tubes for enrichment use.
4 Sterilize using validated laboratory procedures appropriate for the medium formulation.
5 Cool to the recommended inoculation temperature before use.
Note For best isolation performance, GN Broth is commonly used in conjunction with both selective and non-selective agar media.
