Helicobacter pylori is a slow-growing, fastidious, microaerophilic, curved Gram-negative bacterium associated with chronic gastritis, peptic ulcer disease, and other gastroduodenal disorders. Primary isolation from biopsy specimens is often challenging because the organism may be present in low numbers and can be overgrown by contaminating flora if a rich but selective medium is not used.
Helicobacter pylori Selective Medium Base is designed as a nutrient-rich solid support for this workflow. In routine use, the base is prepared with sterile defibrinated blood and a Dent- or VTCA-style selective supplement containing antimicrobial agents that inhibit Gram-positive bacteria, many Gram-negative contaminants, and yeasts or molds while allowing H. pylori to recover under microaerophilic conditions.
The enriched agar base provides peptides, growth nutrients, and osmotic stability; blood contributes additional growth factors and improves recovery of stressed cells; selective supplement limits background organisms; and microaerophilic incubation provides the low-oxygen environment required for colony development.

Prepare the base according to instructions, cool to suitable pouring temperature, then aseptically add sterile blood and the selected Helicobacter supplement.
Streak gastric biopsy or related specimen promptly to minimise viability loss. Fresh biopsy material generally offers the best isolation yield.
Incubate plates under microaerophilic conditions, typically around 35–37°C, for several days. Slow colony development is expected.
Suspect colonies should be confirmed by standard laboratory procedures such as Gram stain, oxidase, catalase, urease, and other appropriate identification methods.
