🧪 Formulation & Selective Profile

📋 Formulation (per Liter)

Component Concentration Role
Proteose Peptone 10.0 g Nutrient base
Yeast Extract 10.0 g B-vitamin source
Sodium Chloride 5.0 g Osmotic balance
Sodium Azide 0.4 g Selective inhibition (G-)
Bromocresol Purple 0.015 g pH indicator
Bacteriological Agar 20.0 g Solidifying agent

Diagnostic Principles

Colony Coloration

Add 1% TTC solution post-sterilization. Enterococci reduce TTC to insoluble red formazan, resulting in deep red colonies.

Acid Production

Fermentation of Maltose and Lactose causes the Bromocresol Purple indicator to shift, aiding in secondary identification.
