🧪 Formulation & Selective Profile
📋 Formulation (per Liter)
| Component | Concentration | Role |
|---|---|---|
| Proteose Peptone | 10.0 g | Nutrient base |
| Yeast Extract | 10.0 g | B-vitamin source |
| Sodium Chloride | 5.0 g | Osmotic balance |
| Sodium Azide | 0.4 g | Selective inhibition (G-) |
| Bromocresol Purple | 0.015 g | pH indicator |
| Bacteriological Agar | 20.0 g | Solidifying agent |
Diagnostic Principles
Colony Coloration
Add 1% TTC solution post-sterilization. Enterococci reduce TTC to insoluble red formazan, resulting in deep red colonies.
Acid Production
Fermentation of Maltose and Lactose causes the Bromocresol Purple indicator to shift, aiding in secondary identification.
