Composition (per litre)

Peptone / Proteose Peptone – 10.0 g
Yeast Extract – 3.0 g
Glucose – 1.0 g
L-Lysine – 5.0 g
Bromcresol Purple – 0.02 g
Agar – 15.0 g

Final pH: 6.8 ± 0.2 at 25 °C


Preparation

Dissolve 34 g in 1 L purified water
Heat until fully dissolved
Autoclave at 121 °C for 15 minutes
Cool to 45–50 °C before pouring


Incubation & Interpretation

Temperature: 35–37 °C
Time: 18–48 hours

Results:
• Purple → Lysine positive (Salmonella likely)
• Yellow → Lysine negative


Storage

Powder: 15–30 °C, dry
Prepared media: 2–8 °C
