Mode of Action
Mebendazole exerts its primary pharmacological action through selective, irreversible binding to β-tubulin at the colchicine-binding site. This binding inhibits tubulin polymerisation into microtubules, disrupting the cytoskeletal architecture required for cell division, intracellular transport, and glucose uptake in susceptible organisms.
Mebendazole binds β-tubulin (colchicine-binding site, irreversible)
↓
Inhibition of tubulin polymerisation into microtubules
↓
Disruption of mitotic spindle & cytoskeletal integrity
↓
Impaired glucose transport → depletion of glycogen stores
↓
Energy depletion → cell death (parasite or target cell model)
Unlike fenbendazole, mebendazole binds tubulin irreversibly and with notably higher selectivity for parasite versus mammalian tubulin isoforms. Mebendazole also exists in three polymorphic forms (A, B, C), with Form C demonstrating the highest biological activity — a factor relevant to formulation and in vitro research design.
