Mode of Action

Mebendazole exerts its primary pharmacological action through selective, irreversible binding to β-tubulin at the colchicine-binding site. This binding inhibits tubulin polymerisation into microtubules, disrupting the cytoskeletal architecture required for cell division, intracellular transport, and glucose uptake in susceptible organisms.

Unlike fenbendazole, mebendazole binds tubulin irreversibly and with notably higher selectivity for parasite versus mammalian tubulin isoforms. Mebendazole also exists in three polymorphic forms (A, B, C), with Form C demonstrating the highest biological activity — a factor relevant to formulation and in vitro research design.
