Description

PPLO Agar is the international reference solid medium for the primary isolation, cultivation, and enumeration of Mycoplasma species (including M. pneumoniae, M. hominis, M. genitalium), Ureaplasma spp., and Acholeplasma laidlawii. It is designed to support the growth of fastidious mycoplasmas from clinical specimens, cell cultures, and pharmaceutical biologics.

Key Characteristics

The medium features an enriched beef heart infusion and proteose peptone base. The very low agar concentration (0.8–1.2%) is critical, as it facilitates the penetration of colonies into the medium, resulting in the characteristic "fried-egg" morphology identifiable under low-power microscopy.

Preparation Method
Base Suspension Suspend 21–25 g in 800 mL purified water. Boil to dissolve.
Sterilization Autoclave at 121°C for 15 minutes.
Enrichment Cool to 50°C. Aseptically add 200 mL sterile serum + 10–20 mL yeast extract.
Additives Glucose/Phenol Red for M. pneumoniae; Penicillin/Ampicillin for bacterial inhibition.
Incubation Parameters

Incubate at 36 ± 1°C in a high-humidity environment for 7–28 days. For M. pneumoniae, an atmosphere of 5–10% CO₂ is essential. Examine plates under a stereo microscope (10–40x) beginning on day 3.
