Rogosa Agar
Overview
Rogosa Agar is a highly selective medium engineered for the isolation and enumeration of Lactobacillus species — especially in dairy products, fermented foods, and probiotic formulations.
Its formulation combines low pH and selective agents (sodium acetate + organic acids) to aggressively suppress competing bacteria, while still supporting the growth of fastidious lactobacilli.

Composition (per litre)
Peptone / Proteose Peptone – 10.0 g
Yeast Extract – 5.0 g
Glucose – 20.0 g
Sodium Acetate – 20.0 g
Agar – 15.0 g
Final pH: 5.4 ± 0.2 at 25 °C
Preparation
Dissolve 70 g in 1 L purified water
Heat with agitation until fully dissolved
Autoclave at 121 °C for 15 minutes
Cool to 45–50 °C before pouring
Incubation & Interpretation
Temperature: 30–37 °C
Time: 48–72 hours (up to 5 days for slow strains)
Results:
• Growth indicates presence of Lactobacillus spp.
• Colony morphology supports species differentiation
