SIM Medium
A comprehensive 3-in-1 differential medium for the simultaneous determination of Hydrogen Sulfide (H₂S) production, Indole formation, and bacterial Motility.
Cat No: AS-1347
ISO 6579 Compliant
FDA-BAM Validated
3-in-1 Diagnostic
Overview & Diagnostic Utility
SIM Medium (Sulfide Indole Motility) is a semi-solid diagnostic tool vital for identifying members of the Enterobacteriaceae family. By combining three tests into a single tube, it streamlines workflows for Salmonella and Shigella identification, saving time and resources in clinical and food safety laboratories.
H₂S PRODUCTION
INDOLE FORMATION
MOTILITY
Key Features
- Motility Detection: Reduced agar concentration (0.35%) allows motile organisms to move through the medium, creating visible turbidity.
- H₂S Sensitivity: Ferrous ammonium sulfate provides a highly sensitive indicator for black precipitate formation.
- Tryptophan Rich: Optimized peptone content ensures reliable indole testing with Kovac’s reagent.

Identification Matrix: SIM Results
| Organism | Sulfide (H₂S) | Indole | Motility |
|---|---|---|---|
| Escherichia coli | Negative (-) | Positive (+) | Positive (+) |
| Salmonella Typhi | Positive (+) | Negative (-) | Positive (+) |
| Shigella dysenteriae | Negative (-) | Varies (+/-) | Negative (-) |
| Proteus mirabilis | Positive (+) | Negative (-) | Positive (+) |
Practical Evaluation
Pros
- Resource Efficient: Replaces three separate tubes with one, reducing incubator space and plastic waste.
- Clear Motility: Much easier to interpret than standard broth or full-agar slants.
- Standardized: Compatible with API 20E systems and international standards (ISO/USP).
Cons
- Technique Sensitive: Requires a perfect vertical stab; sideways movement can mimic false motility.
- Reagent Required: Indole results are not visible until Kovac's or Ehrlich's reagent is added post-incubation.
