Overview

SPS Agar (Sulfite Polymyxin Sulfadiazine Agar) is a selective medium designed for the isolation, enumeration, and differentiation of Clostridium perfringens from food, water, and environmental samples. The combination of sulfite, polymyxin B, and sulfadiazine selectively inhibits the growth of competing bacteria while promoting the development of C. perfringens, which reduces sulfite to form black colonies for easy visual detection.

SPS Agar is widely used in food safety testing, environmental monitoring, and microbiological quality control where selective recovery of sulfite-reducing anaerobes is required.

Preparation
Step Procedure
1 Suspend the medium in purified water and heat to dissolve completely.
2 Dispense into plates or tubes as required.
3 Sterilize by autoclaving at 121 °C for 15 minutes.
4 Cool and pour plates or allow tubes to solidify before use.
