Overview
Thioglycollate Medium, Brewer Modified is an enriched, reducing culture medium designed for the cultivation of obligate anaerobes, microaerophiles, and facultative anaerobes. The Brewer modification provides a dual reducing system — sodium thioglycollate and L-cystine — creating a sufficiently low oxidation-reduction potential to support strict anaerobes without requiring anaerobic jars in routine tube culture.
The medium incorporates resazurin as a visual redox indicator, allowing real-time monitoring of oxygen penetration depth. The semi-solid agar content (0.5 g/L) retards convection and maintains an oxygen gradient. Meets requirements for Fluid Thioglycollate Medium per USP <71>, BP 2023, and EP 2.6.1.
Principle of the Medium
| Sodium Thioglycollate (0.5 g/L) |
Primary reducing agent — scavenges dissolved oxygen, lowers Eh to support obligate anaerobes |
| L-Cystine (0.5 g/L) |
Secondary reducing agent and sulfur source — enhances reducing capacity |
| Resazurin (0.001 g/L) |
Redox indicator — colourless (anaerobic) ↔ pink (oxygen penetration) |
| Agar (0.5 g/L) |
Semi-solid — retards convection, maintains oxygen gradient for growth zone interpretation |
| Casein Digest + Yeast Extract |
Rich nitrogen, vitamins, and growth factors for fastidious anaerobes |
| Dextrose (2.5 g/L) |
Fermentable carbon source and mild reductant |

Key Features
- Dual reducing system — sodium thioglycollate + L-cystine for robust anaerobic environment
- Resazurin redox indicator — visual, real-time monitoring of oxygen penetration
- Pharmacopoeial-grade — meets USP <71>, BP 2023, EP 2.6.1 requirements
- Semi-solid agar — maintains oxygen gradient for growth pattern interpretation
- No anaerobic jar required for routine tube culture
- Nutrient-rich — supports fastidious obligate anaerobes and microaerophiles
Applications
-
Pharmaceutical sterility testing: USP <71>, BP 2023, EP 2.6.1 — parenteral products, biologics, medical devices
-
Clinical microbiology: Cultivation of obligate anaerobes from blood, wound, and abscess specimens
-
Research: Anaerobic physiology, environmental anaerobes, fermentation studies
Preparation
- Dissolve 28.5 g in 1 litre of purified water with gentle heating and agitation.
- Dispense into tubes or bottles as required.
- Autoclave at 121 °C for 15 minutes.
- Cool to room temperature, minimising oxygen exposure.
- Before use: heat in boiling water bath 10–15 min, cool rapidly without shaking.
- Discard any tube where >1/3 of depth is pink (over-oxidised).
CRITICAL: For pharmaceutical sterility testing — use only medium that is colourless or at most faintly pink in the upper layer (≤1/3 depth). Follow full pharmacopoeial preparation and validation requirements.

Key Limitations
- Presumptive medium — confirmatory subculture required for organism identification
- Resazurin pink layer must not exceed 1/3 of tube depth before inoculation
- For pharmacopoeial sterility testing — must be prepared and validated per applicable pharmacopoeia
- Do not overheat — degrades reducing agents and nutrients
