ISO 6579-1:2017 calls for streaking each enrichment broth onto XLD plus a second selective/differential agar chosen by the laboratory. Brilliant Green Agar (BGA) is one of the most widely used choices for that second medium — strong inhibition of E. coli/Proteus/Pseudomonas via brilliant green dye and phenol red, giving pink-red Salmonella colonies.
Confirms presumptive colonies from XLD/BGA via glucose/lactose/sucrose fermentation and H2S production pattern, ahead of further biochemical or serological confirmation. Formulated to ISO 6579 / FDA-BAM standards.
High-buffer Stuart's formulation — rules out urease-positive Proteus spp., which can otherwise mimic Salmonella on TSI (H2S-positive, non-lactose fermenting). Salmonella is urease-negative.

Q1: What does this set cover, and what doesn't it include?
This set covers the culture-media stages of Salmonella detection: pre-enrichment (BPW), dual selective enrichment (RVS and MKTTn run in parallel), and selective/differential plating (XLD). It does not by itself constitute the complete ISO 6579-1 testing and confirmation procedure. MKTTn also requires two additives not included in this set — Lugol's Solution and Novobiocin, both above. Confirmed identification of presumptive colonies requires further biochemical testing (e.g. TSI Agar, Urea Broth) and, where required, serological confirmation — see the complementary media above.
Q2: Why does the workflow use two selective enrichment broths (RVS and MKTTn) instead of one?
RVS Broth and MKTTn Broth have different selective chemistries — RVS relies on magnesium chloride and malachite green at 41.5 °C, while MKTTn relies on tetrathionate, iodine and novobiocin at 37 °C. Different Salmonella serovars and matrices respond better to one broth than the other, so running both in parallel from the same BPW pre-enrichment improves overall recovery.
Q3: What incubation conditions does each medium require?
BPW: 37 °C ± 1 °C for 16–20 hours (pre-enrichment). RVS Broth: 41.5 °C ± 1 °C for 24 ± 3 hours. MKTTn Broth: 37 °C ± 1 °C for 24 ± 3 hours. XLD Agar plates: 37 °C ± 1 °C for 24 hours, read for typical red/pink colonies with black (H2S-positive) centres. ISO 6579-1:2017/Amd 1:2020 broadened several incubation ranges to 34–38 °C — always confirm the exact parameters against your accredited method or the current standard before reporting results.
Q4: Should I plate onto a second selective agar as well as XLD?
Yes — ISO 6579-1:2017 requires streaking each enrichment broth onto XLD plus a second selective/differential agar chosen by the laboratory (not specified by name in the standard). Brilliant Green Agar (BGA) is one of the most commonly used choices and is available as a complementary medium above; Bismuth Sulphite Agar and chromogenic Salmonella agars are other options some laboratories use instead.
