Tryptic Soy Agar (TSA), also known as CASO Agar, is a nutrient-rich, general-purpose solid culture medium widely used for the cultivation and isolation of a broad range of microorganisms in research, quality control, environmental, and industrial microbiology laboratories.
Its balanced formulation, based on enzymatic digests of casein and soybean meal, provides essential amino acids, peptides, vitamins, and minerals that support the growth of many bacteria, yeasts, and moulds. Preparation
-
Suspend 40 g of dehydrated medium in 1 L of distilled or deionised water
-
Heat gently with agitation until completely dissolved
-
Sterilise by autoclaving at 121 °C for 15 minutes

Cool to 45–50 °C

Aseptically dispense into sterile Petri dishes

Allow agar to solidify at room temperature
Storage & Stability

Dehydrated medium: Store at 15–25 °C, dry, in a tightly sealed container

Prepared plates: Store at 2–8 °C, protected from dehydration
